Dengue virus (DENV) antiviral compound testing

Description

This project will involve testing of 15 small molecule drug-like compounds for antiviral activity against dengue virus (DENV-2). Vero cells will infected with DENV-2 (for 3 hours), washed and treated (in triplicate) with each of the 15 test compounds at a range of 8 different concentrations for 72 hours. Cell culture supernatants will then be collected, stored at -80’C and subsequently tested for infectious virus levels by viral plaque assays (in Vero cells). Viral plaques will be enumerated and infectious virus titres calculated. Data will then be processed and graphed (4 parameter dose-response curves [4PL]) to determine antiviral IC50 values for each compound. The results will be compiled and prepared into a report.
